Investigation of nuclear DNA content and cell cycle phases in rat liver cells under chlorpromazine administration

Hepatotoxicity of antipsychotic drugs remains an urgent problem modern medicine. Therefore, the purpose study was to investigate nuclear DNA content and cell cycle phases rat liver cells under Сhlorpromazine administration at doses ranging from 3.5 mg/kg 28 for 30 60 days. The conducted on sexually mature female rats. Chlorpromazine administered once daily days mg/kg, 7 14 21 mg/kg. in nuclei determined by flow cytometry. Cytological analysis performed using FloMax software (Partec, Germany), where percentage G0G1 interval cycle, S phase, G2M interval, apoptosis index – SUB-G0G1 area histograms were determined. Statistical processing results Mann-Whitney U test. showed that has a dose-dependent hepatotoxic effect: with increase dose this drug rats fragmented tissue significantly increased, which is sign hepatocyte death apoptosis. It found did not apoptosis, while highest hepatotoxicity, increasing level 1.9 2.1 (p˂0.05) times, respectively. effect enhanced use days, manifested significant fragmentation, which, our opinion, should be taken into account when conducting long-term therapy patients.
